Compound 33 is a potent 5-HT6 receptor inverse agonist (Ki=23 nM, Kb=6.62 nM). It inhibits downstream Cdk5 and mTOR signaling in vitro and demonstrates in vivo efficacy by alleviating tactile allodynia in a rat model of neuropathic pain following spinal nerve ligation.
