ELISA plate pre-coated by 2 μg/ml (100 μL/well) Human CD123, hFc-His tagged protein (orb689364) can bind Anti-CD123 Neutralizing antibody in a linear range of 0.98-26.70 ng/ml. In order to specifically detect orb689308, mouse anti-human Fab-specific antibody was used as detection antibody.
HEK293 cell line transfected with irrelevant protein (B) and human CD123 (A) were surface stained with anti-CD123 neutralizing antibody 1 μg/ml (talacotuzumab) followed by Alexa 488-conjugated anti-human IgG secondary antibody.
Flow cytometry analysis of antigen binding of anti-human CD123 mAb (orb689308). (A) A clear peak shift of orb689308 was seen compared to the control when incubated with CD123-expressing 8226 cells, indicating strong binding of orb689308 to CD123. Antibodies were incubated at 2 μg/mL.
