Cell Research
I. Nuclear staining 1. Material preparation: (1) Hoechst 33342 solution: Dissolve Hoechst 33342 in an appropriate solvent, such as water or PBS, with a common concentration of 1-10 µg/mL. (2) Cell Samples: Live cells or fixed cells, placed on a slide or in culture. (3) PBS buffer: Used to wash cells and adjust pH. 2. Experimental steps: Live cell staining 1) Add Hoechst 33342 to the cell culture medium to reac the recommended final concentration 2) Incubate at room temperature or 37°C for 15-30 minutes, depending o the cell type. 3) Was the cells with PBS buffer to remove excess dye. 4) Analyz the stained cells using a fluorescence microscope or flow cytometer. Excitation wavelength: 350 nM Emission wavelength: 461 nM Fixed cell staining: 1) Fi the cells using an appropriate fixative (such as 4% paraformaldehyde). 2) After fixation, stai the cells as described above, either on a slide or in suspension. 3) Analyz the fluorescence o the cells using a microscope. II. Application of Cell Cycle Analysis 1. Stain apoptotic cells as described above. 2. Examin the cells for morphological changes, such as chromatin condensation and nuclear fragmentation, which are hallmarks of apoptosis.
