Trastuzumab Biosimilar Antibody, Research Grade (Anti-Her2 / ErbB2) is a research-use monoclonal antibody designed to bind human Her2, also known as ErbB2, CD340 or HER-2/neu. Her2/ErbB2 is a receptor tyrosine kinase and an important oncology research target, particularly in studies of breast cancer, gastric cancer and other Her2-positive tumour models. This antibody is based on the binding specificity of trastuzumab and is intended for laboratory research applications involving Her2/ErbB2 detection, assay development, antibody comparison, biosimilar research and related in vitro studies. It may be useful as a reference-style anti-Her2 antibody in workflows such as ELISA, flow cytometry or other validated immunoassay formats, depending on the user’s own optimisation and experimental conditions. ELISA plate pre-coated by 1 μg/mL (100 μL/well) Human Her2, His tagged protein (orb689435) can bind Anti-Her2 (trastuzumab biosimilar) mAb (orb1173652) in a linear range of 3.2-400 ng/ml.

ELISA plate pre-coated by 1 μg/mL (100 μL/well) Human Her2 Protein, hFc Tag (orb1173800) can bind Anti-Her2 (trastuzumab biosimilar) mAb (orb1173652) in a linear range of 3.81-1730.10 ng/ml. In order to specifically detect orb1173652, mouse anti-human Fab-specific antibody was used as detection antibody.

Flow cytometry analysis with 15 μg/mL Anti-Her2 (trastuzumab biosimilar) mAb (orb1173652) on Expi293 cells transfected with Human Her2 protein (Blue histogram) or Expi293 transfected with irrelevant protein (Red histogram).

Flow cytometry analysis of antigen binding of anti-human Her2 mAb (orb1173652). (A) orb1173652 does not bind to Jurkat cells that do not express Her2. (B) A clear peak shift of orb1173652 was seen compared to the control when incubated with Her2-expressing MCF-7 cells, indicating strong binding of orb1173652 to Her2. Antibodies were incubated at 2 μg/mL.
