| Immunogen | BALB/c mice were immunized with human T lymphoblasts and spleen cells of the immunized animals were fused with X63-Ag8.653 mouse myeloma cells. The supernatants of the resultant hybridomas were screened by a proliferation inhibition assay (inhibition of the human T lymphoblast response to IL-2). Two of those (the clones AHT54 and AHT107) that produced antibodies which inhibited IL-2 dependent proliferation of human T lymphoblasts were selected, cloned and recloned. |

Flow cytometry using the Anti-IL2R antibody AHT54. Paraformaldehyde fixed Jurkat cells permeabilized with 0.5% Triton were stained with anti-unknown specificity antibody (orb256458; isotype control, black line) or the rabbit IgG version of AHT54 (orb1152497, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.
