Storage & Handling
−| Storage | Store vial at -20° C or below prior to opening. This vial contains a relatively low volume of reagent (25 µL). To minimize loss of volume dilute 1:10 by adding 225 µL of the buffer stated above directly to the vial. Recap, mix thoroughly and briefly centrifuge to collect the volume at the bottom of the vial. Use this intermediate dilution when calculating final dilutions as recommended below. Store the vial at -20°C or below after dilution. Avoid cycles of freezing and thawing. |

Biorbyt Anti IL-10 whole antiserum (orb750686) was tested against recombinant IL10. Recombinant IL-10 was run on a 4-20% reducing gel, and transferred to nitrocellulose. Membrane was blocked in 3% BSA-TBS for 1 Hour 4°C and probed with (p/n orb750686) diluted 1:200 in 3% BSA-TBS o/n 4°C. Primary antibody was detected with HRP Gt anti-Rb (orb347654) 1:20000 in orb348637 1 Hour 4°C using FemtoMax Super sensitive chemiluminescent substrate.
Western Blot of Rabbit anti-IL-10 antibody. Lane 1: Recombinant Human IL-10. Load: 50 ng per lane. Primary antibody: IL-10 antibody at 1:500 for overnight at 4°C. Secondary antibody: Peroxidase rabbit secondary antibody (p/n orb347654) at 1:40000 for 30 min at RT. Block: Blocking Buffer for Fluorescent Western Blotting (orb348637) for 30 min at RT. Predicted/Observed size: 17 kDa, 17 kDa for IL-10.
